What the name means

“Semax” is a coined laboratory name rather than an acronym with an agreed public decoding.

What it does encode is chemical lineage: the molecule is built around the fourth-through-seventh amino acids of adrenocorticotropic hormone (ACTH), so it is frequently written as an ACTH(4–7) analog with a short stabilising tail attached.

The commonly reported sequence is Met-Glu-His-Phe-Pro-Gly-Pro (MEHFPGP): the ACTH(4–7) core Met-Glu-His-Phe followed by a Pro-Gly-Pro tail. Reference databases list a molecular formula of C₃₇H₅₁N₉O₁₀S (the sulfur comes from the methionine) and a mass near 814 g/mol.

Where it came from

Semax was developed in the Soviet Union and later Russia, and is commonly attributed to researchers associated with the Institute of Molecular Genetics of the Russian Academy of Sciences and Moscow State University.

It grew out of a long line of Soviet-era work on ACTH-derived peptides and their effects on learning and attention. It has been registered for clinical use inside Russia, which is an important context point: its footprint is largely regional rather than reflecting broad international approval.

Why it was created — the thought process

The starting observation was that fragments of ACTH could influence attention and memory in animal studies without driving the hormone’s downstream steroid effects.

The natural neuroactive fragment ACTH(4–10) is broken down quickly by peptidases. The design logic was to keep the smallest active core (residues 4–7) and bolt on a Pro-Gly-Pro tail — a motif chosen to slow enzymatic degradation and lengthen the peptide’s working lifetime.

How it is made

As a short peptide, Semax is produced by solid-phase peptide synthesis (SPPS).

The chain is assembled one amino acid at a time on a resin, cleaved, then purified by high-performance liquid chromatography (HPLC) with identity confirmed by mass spectrometry. In the Russian research and clinical setting it is commonly prepared as an intranasal solution.

What the research actually shows — honestly

Most of the human data on Semax comes from Russian studies and is not widely replicated by independent groups internationally.

Mechanistic work — reported effects on BDNF expression and on dopaminergic and serotonergic signalling — is largely preclinical and rodent-based. Semax is used within Russia but has not been approved by the FDA or EMA, and large independent trials establishing efficacy and long-term safety are lacking.
