DSIP (Delta Sleep-Inducing Peptide)
A naturally occurring nine-amino-acid peptide first isolated from the blood of sleeping rabbits and named for an EEG effect, with a still-uncertain physiological role.
What the name means
DSIP stands for Delta Sleep-Inducing Peptide — the name is a description of the effect it was first associated with, not its chemistry.
“Delta” refers to delta waves, the slow, high-amplitude brain waves of deep (slow-wave) sleep. The peptide got its name because infusing it was reported to increase delta-wave EEG activity in animals. As section 06 notes, that headline effect has proven far less consistent than the name suggests.
What it actually is
DSIP is a naturally occurring nonapeptide — nine amino acids joined by eight peptide bonds in a single linear chain.
The established sequence is Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu (WAGGDASGE). Reference databases give a molecular formula of C₃₅H₄₈N₁₀O₁₅ and a mass near 849 g/mol.
Where it came from
DSIP was isolated in the 1970s by Schoenenberger and Monnier and colleagues at the University of Bern, Switzerland.
It was recovered from the cerebral venous blood of rabbits whose sleep had been induced by electrical stimulation of the thalamus. That origin story matters: unlike engineered analogs, DSIP is an endogenous substance that researchers pulled out of a living system and then characterised.
Why it was created — the thought process
DSIP was not designed — it was hunted for. The rationale was the search for a circulating “sleep factor.”
The working hypothesis was that sleep might be driven partly by humoral (blood-borne) signalling molecules. By collecting blood from sleeping animals and testing whether it could transfer sleep-promoting activity, the Bern group isolated a candidate substance and named it for its delta-EEG effect.
How it is made
Although DSIP occurs naturally, research and supply-grade material is chemically synthesised.
Being a short peptide, it is made by solid-phase peptide synthesis (SPPS), purified by HPLC and confirmed by mass spectrometry — the same route used for the engineered peptides in this log.
What the research actually shows — honestly
Despite the confident name, DSIP’s sleep-inducing effect has been inconsistent and genuinely controversial across the literature.
Different laboratories have reported effects on sleep, stress responses and pain, but results have been difficult to reproduce reliably, and the peptide’s true endogenous role, receptor and mechanism remain poorly defined. There is no approved therapeutic use.
